The mouse monoclonal antibody 41A recognizes TdT (terminal deoxynucleotidyl transferase; an intracellular antigen), which is responsible for antigen receptor diversity in pre-B and pre-T lymphocytes.
Application
Application details
Flow cytometry: The reagent is designed for intracellular analysis of human blood cells after XPerm (fixation and permeabilization reagent, ED7397) permeabilization of cell membrane and using 10 μl reagent / 100 μl of whole blood or 106 cells in a suspension. The content of a vial (1 ml) is sufficient for 100 tests.

Preparation
Purified antibody is conjugated with R-phycoerythrin (PE) under optimum conditions. Unconjugated antibody and free fluorochrome are removed by size-exclusion chromatography.

Antigen description
Terminal deoxynucleotidyl transferase (TdT) is a template-independent DNA polymerase that catalyzes the addition of deoxynucleotides to the 3'-hydroxyl terminus of oligonucleotide primers. It is expressed in restricted populations of normal and malignant pre-B and pre-T lymphocytes during early differentiation, where it generates antigen receptor diversity by synthesizing non-germ line elements (N-regions) at the junctions of rearranged Ig heavy chain and T cell receptor gene segments. Alternatively spliced transcript variants encoding different isoforms of this protein have been described.
