Antigen description
PCNA (proliferating cell nuclear antigen) is the DNA polymerase delta auxiliary protein acting in homotrimeric form to increase the processivity of leading strand synthesis during DNA replication. PCNA is expressed in the nucleus of all proliferating cells. In response to DNA damage, it is ubiquitinated and is involved in the RAD6-dependent DNA repair pathway. PCNA is a useful marker of DNA synthesis, as its form not involved in DNA synthesis degradates in histological preparations in the presence of organic solvents.

Separation of K562 cells stained using anti-PCNA (PC10) purified antibody (concentration in sample 4 μg/ml, GAM FITC, red-filled) from K562 cells unstained by primary antibody (GAM FITC, black-dashed) in flow cytometry analysis (intracellular staining).
Immunocytochemistry staining of PCNA in PHA-stimulated lymphocytes using mouse monoclonal antibody PC10 (diluted 1:400), detected with GAM IgG-Alexa Fluor®488 (diluted 1:200); DNA stained with propidium iodide. Colocalization of green PCNA signal and red DNA signal indicated as yellow.
Immunohistochemistry staining of human colon (paraffin-embedded sections) with anti-PCNA (PC10), 10 μg/ml. Commercially tested by LifeSpan BioSciences.
