Mouse IgG2a
Specificity
The mouse monoclonal antibody 41A recognizes TdT (terminal deoxynucleotidyl transferase; an intracellular antigen), which is responsible for antigen receptor diversity in pre-B and pre-T lymphocytes.
Application
FC (QC tested)
Application details
Flow cytometry: The reagent is designed for intracellular analysis of human blood cells after XPerm (fixation and permeabilization reagent,
ED7397
) permeabilization of cell membrane and using 4 μl reagent / 100 μl of whole blood or 10
6
cells in a suspension. The content of a vial (0.4 ml) is sufficient for 100 tests.
Reactivity
Human
Immunogen
His-TdT recombinant protein
Preparation
Purified antibody is conjugated with fluorescein isothiocyanate (FITC) under optimum conditions and unconjugated antibody and free fluorochrome are removed by size-exclusion chromatography.
Formulation
Stabilizing phosphate buffered saline (PBS), pH 7.4, 15 mM sodium azide
Storage and handling
Store at 2-8°C. Protect from prolonged exposure to light. Do not freeze.

Antigen description
Terminal deoxynucleotidyl transferase (TdT) is a template-independent DNA polymerase that catalyzes the addition of deoxynucleotides to the 3'-hydroxyl terminus of oligonucleotide primers. It is expressed in restricted populations of normal and malignant pre-B and pre-T lymphocytes during early differentiation, where it generates antigen receptor diversity by synthesizing non-germ line elements (N-regions) at the junctions of rearranged Ig heavy chain and T cell receptor gene segments. Alternatively spliced transcript variants encoding different isoforms of this protein have been described.

Flow cytometry intacellular staining (Excellyse XPerm, cat. no. ED7397) pattern of human peripheral whole blood spiked with REH cellular suspension stained using anti-human TdT (41A) FITC antibody (4 μl reagent / 100 μl of peripheral whole blood spiked with REH cells).
Flow cytometry staining patterns of human peripheral whole blood of acute lymphoblastic leukemia patient.
On left there is surface staining pattern of the sample using anti-human CD45 (MEM-28) PerCP antibody (10 μl reagent / 100 μl of peripheral whole blood).
On right there is intracellular staining (Excellyse XPerm, cat. no. ED7397) pattern of the sample using anti-human TdT (41A) FITC antibody.
Red are TdT positive events - lymphoblastic cells.
Separation of REH cells (red-filled) from human lymphocytes (black-dashed) in flow cytometry analysis (intracellular staining, Excellyse XPerm, cat. no. ED7397) of human peripheral whole blood spiked with REH cellular suspension stained using anti-human TdT (41A) FITC antibody (4 μl reagent / 100 μl of peripheral whole blood spiked with REH cells).
