Specificity
The mouse monoclonal antibody TP25.99SF recognizes an extracellular epitope on HLA-ABC and HLA-E molecules, but not HLA-G. It can be used for discrimination between HLA-G and other HLA-class I antigens.

Antigen description
HLA-class I major histocompatibility (MHC) antigens are intrinsic membrane glycoproteins expressed on nucleated cells and noncovalently associated with an invariant beta2 microglobulin. They carry foreign determinants important for immune recognition by cytotoxic T cells, thus important for anti-viral and anti-tumour defence. Classical human HLA-class I antigens are represented by HLA-A, HLA-B and HLA-C molecules, the non-classical by e.g. HLA-E, HLA-G.

Western blotting analysis was performed on whole cell extracts (RIPA lysis buffer) of JAR, JEG3, Raji, and K562 cell lines, mixed and heated (100°C, 5 min) with non-reducing SDS-loading buffer. Samples were resolved using 10% SDS-PAGE gel.

Nitrocellulose membrane blot was probed with mouse IgG1 monoclonal antibody TP25.99SF (2 µg/ml), followed by IRDye 800CW Goat-anti-Mouse IgG (green). Mouse IgG1 anti-GAPDH monoclonal antibody FF26A conjugated with DyLight 680 (1 µg/ml), was used as the loading control (red). Multiplex fluorescent Western blot detection was performed.

HLA-ABCE molecules were detected at ~40 kDa in JAR and Raji cell lines.
