Isotype
Mouse IgG1 kappa
Specificity
The mouse monoclonal antibody NK92.39 recognizes an extracellular epitope of CD96, a transmembrane glycoprotein expressed at low levels on resting, and at high levels on activated T and NK cells, as well as on many T cell leukemias.
Application
Application details
Flow cytometry: The reagent is designed for analysis of human blood cells using 10 μl reagent / 100 μl of whole blood or 106 cells in a suspension. The content of a vial (1 ml) is sufficient for 100 tests.
Reactivity
Human
Immunogen
human NK92 cell line
Preparation
Purified antibody is conjugated with activated allophycocyanin (APC) under optimum conditions and unconjugated antibody and free fluorochrome are removed by size-exclusion chromatography.
Formulation
Stabilizing phosphate buffered saline (PBS), pH 7.4, 15 mM sodium azide
Storage and handling
Store at 2-8°C. Protect from prolonged exposure to light. Do not freeze.

Antigen description
CD96 is a 160-240 kDa type I transmembrane glycoprotein with intracellular ITIM-like domain, however, without observed inhibitory function. This protein is involved in adhesive interactions of T and NK cells during the late phase of the immune response, and are expressed above all upon their activation. Strong interaction between CD96 and CD155 (poliovirus receptor) stimulates cytotoxicity of activated NK cells, which may play a role in NK cell recognition and killing of CD155-expressing tumor cells. CD96 is expressed on approximately 80% of T cell leukemias and 30% of acute myeloid leukemias and is being used for their immunophenotyping.

Separation of PHA stimulated lymphocytes stained using anti-human CD96 (NK92.39) APC antibody (10 μl reagent per million cells in 100 μl of cell suspension, red-filled) from PHA stimulated lymphocytes stained using mouse IgG1 isotype control (MOPC-21) APC antibody (concentration in sample 5 μg/ml, same as Galectin-3 APC concentration, black-dashed) in flow cytometry analysis (surface staining) of PHA stimulated peripheral blood cells.
