Specificity
The mouse monoclonal HAE-1f recognizes an extracellular epitope of CD62E, a transmembrane glycoprotein expressed on activated endothelial cells, as well as in skin, placenta, and bone marrow endothelium.

Application details
Flow cytometry: The reagent is designed for analysis of human blood cells using 10 μl reagent / 100 μl of whole blood or 106 cells in a suspension. The content of a vial (1 ml) is sufficient for 100 tests.

Preparation
Purified antibody is conjugated with R-phycoerythrin (PE) under optimum conditions. Unconjugated antibody and free fluorochrome are removed by size-exclusion chromatography.

Antigen description
CD62E (E-selectin) is a type I transmembrane glycoprotein of 97 - 115 kDa. It recognizes carbohydrate ligands on various molecules, and promotes leukocyte extravasation by mediating leukocyte rolling on activated endothelia during inflammation. CD62E is expressed above all in inflammated endothelium, but it can also be found on resting endothelium in certain organs, such as skin, or placenta.

Separation of TNF-alpha stimulated HUVEC cells stained using anti-CD62E (HAE-1f) PE antibody (10 μl reagent / 100 μl of cell suspension, red-filled) from HUVEC cells stained using mouse IgG1 isotype control (MOPC-21) PE antibody (concentration in sample 8 μg/ml, same as CD62E PE concentration, black-dashed) in flow cytometry analysis (surface staining) of HUVEC cell suspension.
