Specificity
The mouse monoclonal antibody L161 recognizes an extracellular epitope of CD1c, (R7), a 43 kDa type I glycoprotein associated with beta2-microglobulin. It is expressed on cortical thymocytes (strongly), Langerhans cells, dendritic cells, B and some T cells.

Preparation
Purified antibody is conjugated with R-phycoerythrin (PE) under optimum conditions. Unconjugated antibody and free fluorochrome are removed by size-exclusion chromatography.

Antigen description
CD1c (also known as R7 or BDCA1) together with CD1a and b, belongs to group 1 of CD1 antigens. These non-classical MHC-like glycoproteins serve as antigen-presenting molecules for a subset of T cells that responds to specific lipids and glycolipids found in the cell walls of bacterial pathogens or self-glycolipid antigens such as gangliosides, and they have also roles in antiviral immunity. The trafficking routes of the particular CD1 types differ and correspond to their ability to bind and present different groups of antigens. CD1c is unique in its ability to present e.g. mycobacterial phosphoketides and polyisoprenoids. CD1c is the only CD1 isoform that has been shown to interact both with alpha/beta and gamma/delta T cells.

Anti-human CD1c PE antibody (clone L161) works in flow cytometry application.
Analysis of the antibody staining profile was performed on blood leukocytes isolated from buffy coats. HCDM CDMaps standardized procedures (Kuzilkova D et al. Front Immunol. 2022;13:827898) were used for cell isolation and surface staining of blood leukocytes, with the modification of staining protocol using cytometry test tubes.

HCDM CDMaps standardized procedures (Kuzilkova D et al. Front Immunol. 2022;13:827898) were used for cell isolation and surface staining of blood leukocytes, with the modification of staining protocol using cytometry test tubes.
Suspension of blood leukocytes isolated from buffy coats (2 x 106 cells) was added to the mixture of anti-human CD1c PE antibody (clone L161, 1 µg/ml in stained blood sample) and Monocyte Blocking Buffer (#ED7747), vortexed and incubated for 20 min. Next, optimized backbone antibody panel (HLDA Adaptive) was added to test tubes, vortexed and incubated for 20 min. The residual erythrocytes were lysed with 2 ml of 10× diluted EXCELLYSE Easy solution (#ED7066) and incubated for 10 min. Finally, samples were centrifuged (670 g, 5 min.), supernatant removed and the cell pellet was resuspended in 200 µl of PBS for acquisition.
