Description

Recombinant protein TNF alpha is expressed in S2 cell (Drosophila). DNA sequence encoding 157 AAs was fused with Strep-tag at the N-terminus. A calculated molecular mass of recombinant protein is 18.7 kDa.

Preparation

Purified by affinity chromatography. Lyophilized from a 0.2 μm filtered solution in Storage buffer (100 mM Tris, 150 mM NaCl, 0.01% Tween 20, pH 8.0).

Storage and handling

Store at -20°C to -80°C. Reconstitute in sterile deionized water. Use reconstituted product immediately or aliquot for further storage at -20°C to -80°C.

Recombinant human TNF alpha in functional test

For the test, flow cytometry analysis was performed on HUVEC cells treated or untreated with TNF-alpha. CD54 antibody as surface activation marker was used for activation detection.
Anti-Hu CD54 (MEM-112) purified antibody (concentration in sample 1.5 μg/ml, GAM APC, red-filled histograms) binds specifically to TNF treated HUVEC cells (upper panel), but not to untreated HUVEC cells (lower panel). Level of bacground signal was assesed on cells unstained by primary antibody under same conditions (GAM APC, black-dashed histograms).

Recombinant human TNF alpha purity verification. 5 µg of TNF alpha with > 95 % purity checked by Coomassie Brilliant Blue stained 15% SDS-PAGE.

Recombinant Human TNF alpha induces cytotoxicity in HeLa cell line in the presence of the protein synthesis inhibitor cycloheximide. Measured in WST-8 fluorometric assay method. The ED50 for this effect is 4.2513 ng/mL.

Recombinant Human TNF alpha accelerates apoptosis in HeLa cell line in the presence of the protein synthesis inhibitor cycloheximide. Measured by flow cytometry.
