ELISA: Recombinant protein used as a standard in ELISA may not fully reflect the properties of the native protein, which can lead to inaccurate quantification of its concentration in samples.

Description

Recombinant protein IFN gamma is expressed in S2 cell (Drosophila). DNA sequence encoding 143 AAs was fused with Strep-tag at the N-terminus. A calculated molecular mass of recombinant protein is 18 kDa.

Preparation

Purified by affinity chromatography. Lyophilized from a 0.2 μm filtered solution in Storage buffer (100 mM Tris, 150 mM NaCl, 0.01% Tween 20, pH 8.0).

Storage and handling

Store at -20°C to -80°C. Reconstitute in sterile deionized water. Use reconstituted product immediately or aliquot for further storage at -20°C to -80°C.

Recombinant human Interferon gamma in functional test

We stimulated HeLa cells using recombinant human IFN gamma (according to Blanar MA et al., 1988, PNAS 85: 4672-4676) to induce MHC class II expression on their surface.
Anti-Hu HLA-DR (L243) PE antibody (working concentration 8 μg/ml, red-filled histogram) binds specifically to HLA-DR expressed by IFN gamma treated HeLa cells (upper panel), but not to untreated HeLa cells (lower panel). Level of non-specific binding was assessed using Mouse IgG2a isotype control PE (MOPC-173) under the same conditions (working concentration 8 μg/ml, black-dashed histograms).

Recombinant human IFN gamma purity verification. 5 µg of IFN gamma with > 95 % purity checked by Coomassie Brilliant Blue stained 15% SDS-PAGE.

Recombinant Human IFN gamma induces cytotoxicity in HT-29 human colon adenocarcinoma cell line. Measured in WST-8 fluorometric assay method. The ED50 for this effect is 71.5 ng/ml.
