Description
SingleFlowEx is fluorochrome-labeled monoclonal antibody single reagent for use in laboratory-developed multicolor flow cytometry tests in order to examine human clinical samples.
SingleFlowEx can be used in various multicolor combinations. Laboratory shall establish the SingleFlowEx performance characteristics in conjunction with other reagents in normal and pathological samples.
Contents
1 vial of monoclonal antibody reagent.
The content (1 ml) is sufficient for min. 100 tests.

Stabilizing phosphate buffered saline (PBS), pH 7.4, 15 mM sodium azide
Storage and handling
Store at 2-8°C. Protect from prolonged exposure to light. Do not freeze.

Staining protocol: 10 μl reagent / 100 μl blood sample; intracellular staining
Erythrocyte lysis and permeabilization: EXCELLYSE XPerm (ED7397)
Positive events: Neutrophil granulocytes
Negative events: Lymphocytes
A - Flow cytometry intracellular staining pattern of human peripherial whole blood stained using anti-human Myeloperoxidase (MPO421-8B2) PE antibody (10 μl reagent / 100 μl blood sample). Black - EDTA-anticoagulated human peripheral blood, healthy donor; Blue - Heparin-anticoagulated human peripheral blood, patient with MPO deficiency; Red - Heparin-anticoagulated human peripheral blood, mother of patient with MPO deficiency. B - Separation of human Myeloperoxidase positive neutrophil granulocytes of healthy donor (black-filled), patient with MPO deficiency (blue-filled) and mother of patient with MPO deficiency (red-filled) in flow cytometry analysis (intracellular staining) of human peripheral whole blood stained using anti-human Myeloperoxidase (MPO421-8B2) PE antibody (10 μl reagent / 100 μl of peripheral whole blood).
