CD34 at a glance

CD34, one of the mucosialins, is a highly glycosylated transmembrane phosphoglycoprotein, which has a great importance in selection and enrichment of hematopoietic stem/progenitor cells (HSPC) for bone marrow transplantation. It is expressed on cells from diverse tissues that have comparable properties, such as the progenitor and stem cell activity, as well as the transdifferentiation potential. Namely it is on hematopoietic stem cells (HSC), multipotent mesenchymal stromal cells (MSC), muscle satellite cells, keratocytes, hair follicle epithelial stem cells, vascular endothelial progenitors, and other. On these cells, CD34 is coexpressed with tissue specific markers, and its expression is lost upon activation of differentiation process. CD34 is also being used as one of the markers for staining blasts in acute myeloid leukemia, and it is also expressed in some tumors.

Antibodies specific for CD34 have been classified according to the type of their epitope into several groups:

  • Class I (Ia, Ib): Epitope is sensitive to treatment with neuraminidase and glycoprotease (class Ia: very sensitive, class Ib: partially sensitive).
  • Class II: Epitope is sensitive to glycoprotease, but it is resistant to neuraminidase. Example: clone QBEnd-10 (HLDA workshop V).
  • Class III: Epitope is resistant both to neuraminidase and to glycoprotease (glycosylation-independent epitope). Example: clones 4H11 (HLDA workshop VI), 581 (HLDA workskop V); both clones have comparable flow cytometry staining pattern.

SingleFlowEx CD34 reagents
SingleFlowEx is fluorochrome-labeled monoclonal antibody single reagent for use in laboratory-developed multicolor flow cytometry tests in order to examine human clinical samples.
SingleFlowEx can be used in various multicolor combinations. Laboratory shall establish the SingleFlowEx performance characteristics in conjunction with other reagents in normal and pathological samples.

Fig.2: Western blotting analysis of human CD34 using mouse monoclonal antibody QBEnd-10 on lysates of TF-1 cell line and HEK293T/17 cell line (CD34 non-expressing cell line; negative control) under non-reducing and reducing conditions. Nitrocellulose membrane was probed with 2 µg/ml of mouse anti-CD34 monoclonal antibody QBEnd-10 followed by IRDye800-conjugated anti-mouse secondary antibody. A specific band was detected for CD34 protein at approximately 110 kDa.
