Flow cytometry surface staining patterns of five CD3 antibody clones (OKT3, TB3, SK7, UCHT1, MEM-57) when used simultaneously with TCR alpha beta antibody staining. TCR alpha beta signal is impaired when stained simultaneously with some CD3 clones. This results in insufficient distinction between TCR alpha beta positive and negative CD3 positive T cells. Out of the tested clones, OKT3 interferes with the TCR alpha beta staining the most. On the other side of the spectrum MEM-57 provides the best distinction between positive and negative populations while maintaining MFI of TCR alpha beta single stained control. TB3, SK7 and UCHT1 clones manifest various degrees of blocking as demonstrated in Figure 1. Staining was performed on three different donor samples.
