Mouse monoclonal antibody MAb1 is suitable for Western blotting, ELISA, and neutralization of human TNF alpha. Mouse monoclonal antibody MAb11 is suitable for flow cytometry, immunocytochemistry, immunohistochemistry, ELISA and neutralization of human TNF alpha, and it was reported to crossreact also with monkey and porcine antigen. Comparison of both antibodies concerning applications demonstrates fig. 1. We have used MAb1 in the concentration of 5 µg/ml as the capture antibody, and biotinylated MAb11 in the concentration of 0.15, 0.31, 0.63, 1.25, 2.5, 5, and 10 µg/ml in a sandwich ELISA to detect 0, 0.05, 0.45, and 4.1 ng/ml of human TNF alpha. Results are demonstrated in fig. 2. We were able to detect TNF alpha even in the concentration of 0.05 ng/ml (50 pg/ml) and it was obvious that the detection limit is even lower.
