Background

The oncoprotein ErbB2/HER2 (human epidermal growth factor receptor 2), also known as Neu or CD340, is a 185 kDa transmembrane tyrosine kinase of cell surface growth factor receptor family. It is present in a wide variety cell types of normal human fetal and adult tissues and is frequently overexpressed in human carcinomas (e.g. in 20-30% cases of breast cancer cells). Activation of ErbB2 triggers intracellular signalling events, which are essential for cell growth and differentiation. In the last 20 years ErbB2 antigen has become very important marker and therapy target in patient care.

Specificity

The polyclonal antibody reacts with ErbB2/HER2 oncogen, a 185 kDa transmembrane tyrosine kinase presented in a wide variety cell types of human tissues.

Immunogen

Synthetic peptide corresponding to a site on the extracellular domain of the c-ErbB2 oncoprotein (HER2/neu).

Applications:

  • Immunohistochemistry (paraffin sections)
    Recommended dilution: 10 μg/ml in PBS with 10% FCS and 0.1 % NaN3
    Staining technique: standard ABC technique (DAB+)
    Pretreatment: Fill the staining dishes with 10 mM citrate buffer pH 6.0 (pretreatment solution). Put the staining dishes with the pretreatment solution in the water bath and heat on 95-99°C. Sink the deparaffin sections into the preheated pretreatment solution. Once again heat the water bath and pretreatment solution with sections on 95-99°C - incubate for 40 min in 95-99°C. Remove the dishes with sections from the water bath and keep them cool down for 20 min in RT. Rinse the sections 3 x 5 min in washing buffer (PBS).
    Incubation: overnight at 4°C
    Positive tissue: breast carcinoma
    Application note: Warning: The sections must not drain!!
Usage note:
Indicated dilutions are recommended starting points for use of this product. Working concentrations should be determined by the investigator.
